Association of HLA with Vogt-Koyanagi-Harada Disease in Taiwan |

PURPOSE:
To study the distribution of human leukocyte antigen HLA-A/B antigens and HLA-DR/DQ alleles and to investigate the immunogenetic background of Taiwanese patients with Vogt-Koyanagi-Harada (VKH) disease
METHODS:
Human leukocyte antigen typings were performed in 53 Taiwanese patients with VKH syndrome and in 73 healthy control subjects. HLA-A/B loci serologic typing was performed according to the standard microlymphocytotoxicity technique. DNA was extracted through the salting out method and HLA-DR/DQ phenotyping were performed with the polymerase chain reactionsequence specific oligonucleotide probe (PCR-SSOP) method.
RESULTS:
Among HLA-A/B antigens typed by the standard microlymphocytotoxicity method, the frequencies of HLA-B54 (26.42%, P ≦ 0.001), were significantly increased in the patient group compared with the control group. Among HLA-DR/ -DQ alleles subtyped by DNA methods, the frequencies of HLA-DR4 (84.9%, P ≦0.001) and HLA-DQ4 (77.36%, P ≦0.001) were significantly increased. However, significant decreases in HLA-DR8( 5.66%, P ≦0.025 ), HLA-DR11( 0%, P ≦0.001 ), HLA-DR12( 16.98%, P ≦0.05), HLA-DQ6( 16.98%, P ≦0.025) and HLA-DQ7( 18.87%, P ≦0.01 ) frequencies were observed.
CONCLUSIONS: These results suggest that HLA- B54, HLA-DR4, HLA-DQ4 is greatly increased and may play an important role in the development and the clinical course of VKH disease in Taiwanese patients.
